Asialoglycoprotein receptor (ASGPR) is the ideal target for the delivery of therapeutic oligonucleotides to the liver due to its tissue specificity, high expression levels, rapid internalization, and turnover. N-acetylgalactosamine (GalNAc) is a well-defined liver-targeted moiety benefiting from its high affinity with ASGPR. Oligonucleotides containing GalNAc can be deprotected using standard procedures during which the acetyl protecting groups on the GalNAc group are removed.
